Procedure for reviving cultures from L-Dried materials.
 |
- Prepare the rehydration fluid and the growth medium fit in the cultures.
- Score the tube around at the mid position of cotton plug inserted in the tube.

- Sterilize the surface of the tube with 70% alcohol-dampened gauze. Please take care not to erase strain number written on the surface of the tube.
- Wrap the tube with sterilized layered-gauze and break it gently.

- Take 0.2 to 0.5 ml of rehydration fluid with a Pasteur pipette. Add a tiny drop of fluid the dried material and moisten cells well. Repeat the moistening procedure2 to 3 times. Add fluid remaining in the pipette and suspend cells well.

- Spread 0.1ml of cell suspension onto agar plate medium.
- Inoculate liquid medium in 2 to 3 test tubes with remaining cell suspension.
- After the growth, check the purity of the culture and then use for experiments.
